    摘要:

    菜豆种子粗酶液经乙醇-氯仿、丙酮沉淀、DEAE-纤维素柱层析和Sephadex G-75柱层析,获得比活力为127.45U/mg的超氧化物歧化酶(SOD),该酶活性受H2O2和KCN强烈抑制,经电泳鉴定为3种SOD同工酶,部分纯化的SOD证明为Cu,Zn-SOD,经不连续聚丙烯酰胺凝胶电泳获得其中1种电泳纯的酶,超声波处理此酶后,酶活力发生改变,适宜的超声波参数能显著提高酶活力,经超声波处理后,SOD的紫外吸收光谱无明显变化,而紫外差示光谱在一定波长范围内出现正峰和负峰。

    Abstract:

    Isoenzyme of superoxide dismutase(SOD) from kidney beans (Phaseolus vulgaris . L) seeds was purified by steps involving crude extract, chloroform-ethanol treatment, acetone precipitation, DEAE-cellulose chromatography and Sephadex G-75 chromatography. It had a specific activity of 127.45 U /mg. Its activity was strongly inhibited by H2O2 and KCN. It was proved that the purified enzyme was copper-and zinc-containing SOD. One of the isoenzymes was purified to homogeneity by discontinuous PAGE. The activity of this SOD could be changed by ultrasound treatment. The activity of SOD was increased after it was dealt with by ultrasound of suitable parameters. The ultraviolet absorption spectrum of SOD had no obvious change after treatment by ultrasound. Differential UV-spectra presented distinct positive peaks and negative peaks within certain wavelenth after ultrasound treatment.
